Genistein is a naturally occurring isoflavone found primarily in soybeans and soy-derived products. It belongs to the flavonoid class of polyphenols and is chemically characterized as 5,7-dihydroxy-3-(4-hydroxyphenyl)chromen-4-one. In its natural form, genistein often exists as a glycoside (genistin), which is converted to the aglycone form through hydrolysis during processing or fermentation. The genistein content is determined by high-performance liquid chromatography (HPLC) using an external standard method. A C18 reversed-phase column is employed with a mobile phase consisting of methanol/water (or acetonitrile/water) with a small amount of acid. Detection is carried out at a UV wavelength of approximately 260 nm. The sample is dissolved in methanol, filtered, and injected; quantification is performed against a certified genistein reference standard. Results are calculated on a dried basis.
